how to subtract 2 elements in photshop when they are in two layers

all my question is how can i delete the intersection of two layers i have tried almost every question SO but could not find my solution. i wanted to delete the intersection i m following a video on youtube LINK for Video but i m unable to do that in my photoshop. i tried subtracting type thing,i m able to make that final image using a single layer but i want to do that in between 2 layers.answer i found on SO are all about one layers.
